Rare Cells: This module will focus analysis of rare cells, in general subsest represented below 1% of all cells. Condierationf of sample preparation, acquistition and hardware needs and anaysis will be discussed. Exampl eof arare cells are stem cells as well as antgen specfiic T-cells.
Mitochondria: The wide range of probes available has expanded the chance to characterize in deep phenotype and functionality of mitochondria, powerhouse of the cells. However, this has also increased complexity of using fluorochrome combination due to spectral overlap and need for multilaser flow cytometry. In this wet lab, we will focus on different probes used, the correct use of positive and negative controls in this analysis.  A complex application of the theories behind these approaches will be presented. In particular, the wet lab will be focused on the assessment mitochondrial mass, mitochondrial membrane potential and reactive oxygen species.

This module focuses on the basic principles of flow cytometry such as light, fluorescence, excitation and emission, cytometry instrumentation (fluidics, optics, electronics) etc.. It will also cover multicolor panel design with information on different fluorochomes, spectral spillover and compensation. Importantly, cytometer variables affecting measurements will be discussed as well.

This module will explain principles of particle sorting, discuss flow sorter setup and sample preparation.

This module will give detailed protocol on sample fixation and permeabilization for flow cytometry of surface and intracelular markers. Moreover, discussion over different effects of detergents on cells and epitopes, inculding the effects of different concentrations and incubation times will be given. The module will be complemented by a practical part full of usefull lab tips.

This module will present practical usage of data reduction tools (PCA, ViSNE and UMAP), will introduce hints on how to work with data cohorts.

This practical module will guide you through the jungle of various sample preparation techniques, starting from lyse no wash protocol to arrive at staining of intracellular cytokines. Participants will learn how to prepare cells for basic surface staining, how to freeze and store cells for later staining and how to stain cells for detection of intracellular markers.
